Understanding CRISPR: A Primer
CRISPR-Cas9 has transformed the landscape of molecular biology by providing a precise, programmable tool for editing DNA. This article offers a brief primer for students new to the technology.
How it works
CRISPR systems use a short guide RNA to direct the Cas9 enzyme to a specific location in the genome, where it introduces a double-strand break. The cell’s own repair machinery then fixes the break, allowing researchers to disrupt, correct, or insert genetic sequences with high precision.
Applications
- Functional genomics — studying gene function by targeted knockout
- Disease modeling — creating cell and animal models that carry disease-associated mutations
- Therapeutic development — correcting disease-causing mutations directly in patient cells
Members of our department use CRISPR-based approaches across several research programs, from cancer cell biology to microbial genome engineering.
